Abstract
The interactions of antiseptic compounds with quaternary ammonium, such as benzalkonium chloride (BC), benzethonium chloride (BZC), dodecyldiaminoethyl-glycine hydrochloride (AEG), and chlorhexidine gluconate (CHG), with components of the stratum corneum were investigated by isothermal titration calorimetry at pH 7.5 and 25 °C. The different mechanisms for their permeation to stratum corneum were clarified. Cationic surfactants of BC and BZC bound to cholesterol and cholesterol sulfate with high affinity (105–106 M−1) to extract endogenous cholesterol and its derivatives from the stratum corneum and penetrated via an intercellular route. CHG also bound to cholesterol and accumulated in the stratum corneum without removing endogenous cholesterol. On the other hand, an amphoteric surfactant of AEG seemed to be incorporated into the lipid bilayer and bound to ceramide with its polar end close to the lipid polar heads by hydrophobic interaction.
