  • Abstract
    Résumé epteur à l’EGF (EGFR) est un récepteur membranaire, surexprimé dans de nombreux cancers épithéliaux. Son activation déclenche plusieurs voies de transduction du signal, avec pour conséquences une induction de la prolifération cellulaire, une inhibition de l’apoptose et une néoangiogenèse. L’utilisation d’anticorps monoclonaux (cetuximab) ou d’inhibiteurs de la fonction tyrosine-kinase du récepteur permet d’inhiber ces phénomènes. En modulant ces fonctions cellulaires, ces molécules possèdent une action radiosensibilisante. Plusieurs mécanismes ont été évoqués (altération des réparations des lésions radio-induites, facilitation de l’apoptose, action sur la repopulation en cours de traitement) et justifient les essais cliniques d’association actuellement en cours. ceptor (EGFR) is a transmembrane protein, which was implicated in the progression of many epithelial cancer types. Its activation induce some transduction pathways inside the cell, and contribute to many cellular processes as cell proliferation, inhibition of apoptosis and angiogenesis. Monoclonal antibodies directed against EGFR or small molecules inhibiting its tyrosine-kinase function could block all these pathways. By modulating these cellular functions, these molecules enhance the antitumor activity of ionizing radiation. Several mechanisms have been discusssed (alteration od DNA damage repair, faciliation of apoptosis, inhibition of tumour repopulation) and justifie the current clinical trials combining these drugs and radiotherapy.
