  • Abstract
    The purpose of this work was to examine the allele structures of beta-lactoglobulin gene with restriction fragment length polymorphism (RFLP) method in Simmental, Brown Swiss, South Anatolian Red and Turkish Grey cattle breeds being raised in Turkey. The material of the study consisted of 75 heads of Simmental, 75 heads of Brown Swiss, 40 heads South Anatolian Red and 40 heads Turkish Grey cattle. DNA materials were isolated from blood samples using phenol-chloroform extraction method. In order to determine the beta-lactoglobulin alleles in polymerase chain reaction (PCR) products, the PCR products were digested with HaeIII endonuclease enzyme. The digest products obtained were determined by electrophoresis using 4% agarose gel. The AA genotypic frequency was found the highest in Simmental breed; the BB genotypic frequency was found the highest in the South Anatolian Red (SAR) breed and the AB genotypic frequency was found the highest in Brown Swiss breed in beta-lactoglobulin gene. In this study, the B allele frequency was found higher than the A allele in SAR cattle breeds. But, the A allele frequency was found higher than the B allele in the Simmental breed. At the end of this study, a deviation in Hardy-Weinberg equilibrium (HWE) was observed in terms of beta-lactoglobulin locus in the breeds examined.
