Treatment of corneal tranplant rejection in human with anti-interferon-γ antibodie

PURPOE: To determine efficacy of anti-human interferon-γ F(ab′)2 (Fab) in treating corneal tranplant rejection after penetrating keratoplaty. DEIGN: Interventional cae erie. METHOD: Anti-interferon-γ Fab derived from goat antihuman interferon-γ antibodie were ued for intillation in 13 patient (13 eye) who experienced corneal tranplant rejection after penetrating keratoplaty. Fab were adminitered in one or three treatment coure() a eyedrop given three time daily for 7 to 10 day. REULT: In 10 patient, 2 to 3 day after tart of one treatment coure, tranplant tranparency improved, and edema decreaed. At the end of week 1, the tranplant became almot fully tranparent, and eye inflammation diappeared. Viual acuity increaed from 0.2 to 0.3. In the other three patient, three treatment coure were needed for comparable improvement. Improvement wa maintained through follow-up—an average of 7 month. CONCLUION: Antihuman interferon-γ Fab may be afe and effective in halting corneal tranplant rejection after penetrating keratoplaty
