  • Abstract
    Purpoe To determine the interreader and intrareader agreement at the Optical Coherence Tomography (OCT) Reading Center at Duke for image produced for an interventional neovacular age-related macular degeneration (AMD) clinical trial. Deign Retropective, obervational cae erie. Method OCT wa performed uing the tratu OCT Fat Macular Thickne Map (Carl Zei Meditec, Dublin, California, UA) can mode and a 7-mm line can centered on the fovea. Experienced OCT reader, certified to grade can according to a tandardized protocol, independently determined whether a can wa gradable. If gradable, each of the can wa graded for multiple morphologic characteritic. In addition, retinal thickne, ubretinal fluid thickne, and choroidal neovacularization (CNV) thickne at the fovea were meaured for each gradable can. Interreader agreement wa determined among three reader pair. Reader regraded a ubet of can and intrareader agreement wa determined. Reult The interreader agreement wa high for can gradability and can grade among three reader pair, ranging from 84% to 100% and from 84% to 96%, repectively. imilarly, the intrareader agreement for can gradability and can grade, including comparion of adjudicated can grade with readjudicated can grade, wa high and ranged from 91% to 100% and from 79% to 98%, repectively, except for can grade of retinal pigment epithelium atrophy, when CNV wa preent. Highly reproducible reult alo were found for quantitative thickne meaurement. Concluion Well-trained OCT reader can grade independently, with a high level of interreader and intrareader agreement, multiple morphologic parameter of OCT can obtained from eye with neovacular AMD. Reproducible tratu OCT can data will be valuable to monitor treatment efficacy in interventional clinical trial of neovacular AMD.
