امثلة ظروف انتاج انزيم السيلولاز من بكتريا Bacillus cereus 21 باستخدام طريقة التخمير المغمور

The conditions for producing cellulase enzyme were optimized using corn husks as substrate in submerged fermentation. The effects of five parameters (incubation temperature, pH, substrate concentration, inoculum volume, fermentation time) on the strain growth and enzyme production were studied. For this purpose the statistical program Minitab and the statistical design Response Surface Methodology (RSM) were applied. The highest enzyme activity was obtained at an incubation temperature of 25ºC, pH value of 9.5, fermentation time of 18 h, inoculum volume of 1% and substrate concentration of 0.5%. Total enzyme activity was deduced through the amount of reduced sugar produced by extracellular cellulose degrading enzymes, where their concentration is proportional to total activity of cellulase enzymes. Enzyme activity obtained under the above mentioned conditions was 0.6 IU/ml
