    Characterization of Some Certified Pear (Pyrus communis L.) Genotypes With Morphological and RAPD Methods

  • Abstract
    In this study, the genetic relationships among six pear varieties and genotypes intensively cultivated in our country were determined using RAPD (Randomly Amplified Polymorphic DNA) molecular marker technique. Molecular analysis with eight RAPD primers produced 48 polymorphic bands. The highest number of polymorphic bands (11) were obtained from primer OPA-08 while the least number of polymorphic bands (two) were obtained from primer OPE-07. The average number of polymorphic bands per primer was six. Among all varieties, the highest similarity index was determined between Klasik Akça and Kokulu Akça (0,922) but the lowest similarity index was identified between Kokulu Akça and Santa Maria (0,431). The clustering analysis showed that all genotypes were separated into two main groups and each main group was divided into two subgroups. While Klasik Akça and Kokulu Akça genoytpes were clustered into the first subgroup; Deveci pear formed the second and Saman Akça and Dikenli Akça formed the third subgroups. The fourth subgroup contained Santa Maria variety. Moreover, the genotypes were also compared pomologically based on the UPOV criteria. The results showed significant differences among varieties in terms of fruit weight (30,19-305,0), fruit width (3,76-8,15), fruit length (5,89-10,6), fruit shape index (0,94-1,57), fruit firmness (42,03-66,1), fruit peel color (L 64,11-75,81, a 14,55-6,86, b 27,05-50,3), fruit juice pH (2,51-5,16), total soluble solids (8,95-10,4), pedicel length (4,95-2,32), pedicel thickness (2,8-4,67), pedicel bowl width (3,65-13,09), pedicel bowl depth (1,03-4,65), flower bowl width (7,93-13,94), flower bowl depth (3,65-10,74), seed bowl depth (23,86-34,73), seed bowl width (11,59-26,98), core lenght (8,34-9,19), core width (2,8-4,68) and core thickness (1,1-3,18).
