Abstract
We studied the protective effect and antioxidant mechanism of Xiongma pills-containing serum on H2O2 injury in PC12 cells. Oxidative stress models were established by H2O2 pretreated with PC12 cells. Cell viability, lactate dehydrogenase release(LDH) in culture supernatants and glutathione peroxidase (GSH-PX) activities in suspension cells were determined by MTT assay and chemistry determination, respectively. H2O2 could induce PC12 cell damage and decrease survival rate of PC12 cells significantly, increase LDH leakage and decrease GSH-PX activities. Xiongma pills-containing serum which the administration dose was equivalent 0.75, 1.5, 3, 6.g·kg-1 crude drug respectively could significantly increase the survival rate of PC12 cells and GSH-PX activities, and decrease LDH leakage. And Xiongma pills 6 g·kg-1 crude drug was the best dose. Our study suggests that Xiongma pills-containing serum has a neuroprotective effect against H2O2-induced antioxidant through mechanisms related to improve the activity of antioxidative enzymes via and scavenge reactivity oxygen species.
