Abstract
CAPS marker is an efficient method to validate candidate SNP. In this study, we analyzed 42,493 grape EST sequences and 5,032 candidate SNP were identified, the amount of transversions, transitions and one base indel were 1,800, 2,896 and 336 respectively. The CAPS reaction system contained ddH2O 5.5μL, buffer 1μL, restriction enzyme 0.5μL, PCR product 3μL. In this study, 20 grape materials were used for enzyme digestion analysis, the genotype were successfully resolved. CAPS marker is fast, low cost and efficient in grape EST-SNP analysis, and holds great promise for molecular assisted breeding or other researches of grape.
