A microplate technique for phospholipase D activity determination

A new microplate phospholipase D (PLD) assay has been developed on the basis of a two-step technique with colorimetric determination of the reaction product choline. The described method was optimized concerning its linearity, reproducibility and concentrations of reaction components. The results obtained with the microplate technique in the analysis of different PLD samples were compared with those obtained with the biosensor assay. The microplate assay proved to be more sensitive than the biosensor technique and considerably less time-consuming.
