Unraveling the sequence of reactions that transforms aromatic polyketide backbones into much more elaborate structures, for example the chartreusin bislactone, is challenging, but in this issue of Chemistry & Biology, Xu and coworkers [] show that sequencing the gene clusters can provide valuable clues.
