HMGB1 and the recently described interleukin-33 (IL-33) are abundant chromatin-associated nuclear factors with potent proinflammatory cytokine activities. In this issue of Chemistry & Biology, Mollica et al. report the identification of the first direct small-molecule inhibitor of HMGB1.
