Abstract :
آوي باكتريوم (هموفيلوس) پاراگالينارم، باكتري گرم منفي است كه ايجاد بيماري حاد تنفسي كوريزاي عفوني در ماكيان مي كند. كوريزاي عفوني در بيشتر كشور ها ديده مي شود. در گله هاي طيور تخمگذار و مادر در ايران نيز اين بيماري ديده مي شود. اطلاعاتي تاكنون در كشور در خصوص سروتيپ هاي شايع باكتري عامل اين بيماري وجود ندارد. اين مطالعه جهت تعيين سرو وار دو مورد جدايه از قبل گزارش شده اين باكتري به روش واكنش زنجيره اي پليمراز اجرا شد. اين جدايه ها در زرده تخم مرغ فريز شده نگهداري مي شدند كه بطور غير عمدي غير فعال شده بودند. لذا استخراج ژنوم باكتري از زرده به روش فنل – كلروفرم انجام گرفت و سپس محصول هاي نهايي استخراج، تحت آزمايش واكنش زنجيره اي پليمراز با استفاده از پرايمر هاي اختصاصي گروه اوي باكتريوم پاراگالينارم قرار گرفتند. بدنبال آن با مثبت شدن نتايج، مجدداً آزمايش واكنش زنجيره اي پليمراز ديگري با استفاده از پرايمر هاي شناسايي سروار هاي A, B, C بر اساس تفاوت هاي توالي ژني مربوط به پروتيين سطحي هماگلوتينين، انجام شد.
Abstract :
Avibacterium paragallinarum is Gram-negative bacteria that cause infectious coryza (IC), an acute respiratory disease of chickens. Despite vaccination of Layer and breeder farms of Iran against IC, they are still experiencing the disease clinically and there is no knowledge of serotypes prevalence of this bacterium in this country. This study designed to determine serovar identity by molecular identification tests of previously studied isolates from Iran that were inactivated accidentally in egg yolk storing culture media. Genomes of the isolates extracted from egg yolk and then were subjected to two set of PCR tests using by published group specific and Pageʹs serovar specific primers. The later primers were based on sequence differences in hypervariable region of haemagglutinin protein of Av. paragallinarum. These two sequential PCR confirmed that the bacteria were Av. paragallinarum and were associated to Pageʹs serotype B. This is the first report of molecular detection of serotype B of Av. paragallinarum from Iran.
