Abstract :
Heparin/fluorocarbon composite film was prepared by depositing heparin on fluorocarbon using radio frequency (RF) sputtering. The preparation and anti-blood coagulation characterization of heparin/fluorocarbon complex film were investigated in this paper. The preparation of heparin/fluorocarbon composite film included three steps. First, fluorocarbon film was deposited on a substrate target polytetrafluoroethylene (PTFE) using RF sputtering. Second, glycidyl methacrylate (GMA) was grafted on the surface of fluorocarbon film. Finally, the heparin was covalently bonded to GMA. The chemical compositions of the film surface at different preparation stages were investigated by X-ray photoelectron spectrum (XPS). The results of XPS showed that the GMA had been grafted on the surface of the fluorocarbon film and the heparin has been immobilized on the PGMA/fluorocarbon film. The surface morphology was investigated by scanning electron microscope (SEM). The anti-blood coagulation property was determined by Activated Partial Thromboplastine Time (APTT) and Thrombin Time (TT) tests. The results showed that Heparin/fluorocarbon composite film showed well anti-blood coagulation property. The values of APTT and TT increased with increasing immobilization yield of heparin. Compared with the PTFE, the APTT and TT of the heparin/fluorocarbon composite film increased by 1∼2 times. It can be seen that the grafting yields increase with increasing grafting time throughout the reaction.
