Abstract
The study was conducted to develop a micro propagation system for Dracocephalum Kotschyi, an important-rare medicinal plant in Iran. MS media containing Zeatin or KIN (0, 1 and 2 mg.l-1) in combination with IBA and NAA (0, 0.2 and 0.5 mg.l-1) were investigated for shoot regeneration using nodal explants. The average length of microshoots was obtained on KIN (2 mg.l-1) alone or combined with 0.5 mg. l-1NAA or IBA. A maximum lateral shoots were produced on IBA at 0.2 mg.l-1 or KIN 1 mg.l-1.No sign of rooting was observed on treatments containing ZEA.Percentage of rooted microshoots reached to highest in some treatments such as KIN (1 mg.l-1) + NAA (0.2mg.l-1). Well-developed shoots with strong root were transplanted to the pots filled with a mixtureof peat + cocopeat+ perlite (2:1:1) under controlled greenhouse condition. All plantlets were survived and showed normal growth.
