Abstract :
Objective- Adipose tissue is an appropriate source for isolation of cells with stem-cell–like properties. In the present long-term study, the effects of the omental adipose-derived nucleated cells (OADNCs) fraction were compared to those of the undifferentiated cultured bone marrow stromal cells (BMSCs) on sciatic nerve regeneration. Design- Experimental in vivo study. Animals- Fifty male White Albino rats. Procedures- The rats were divided into four experimental groups, randomly: Normal control (NC), inside-out vein graft (IOVG), the OADNC and the BMSC groups. A 10-mm sciatic nerve defect was bridged using an inside-out vein graft. In OADNC and BMSC groups, the vein was filled with OADNCs and BMSCs, respectively. Functional studies of sciatic function index, electrophysiologic, morphometric and immunohistochemistry assessments were performed at 16, 20 and 24 weeks after surgery. Results- There was no significant differences (P > 0.05) between BMSC and OADNC groups concerning recovery of the regenerated nerves, amplitude and time delay of electromyography. Compared to IOVG, OADNCs enhanced the nerve regeneration similar to undifferentiated BMSCs. Conclusion and Clinical Relevance- It was concluded the long-term morphometric results could be more reliable in histological study of sciatic nerve regeneration.

ﻫﺪف- ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﻳﻚ ﻣﻨﺒﻊ ﻣﻨﺎﺳﺐ ﺑﺮاي اﺳﺘﺤﺼﺎل ﺳﻠﻮلﻫﺎي ﺑﺎ وﻳﮋﮔﻲﻫﺎي ﺳﻠﻮلﻫﺎي ﺑﻨﻴﺎدي ﻣﻲﺑﺎﺷﺪ. در ﻣﻄﺎﻟﻌﻪ ﺣﺎﺿﺮ ﺗﺎﺛﻴﺮ ﻃﻮﻻﻧﻲ- ﻣﺪت ﺳﻠﻮلﻫﺎي ﻫﺴﺘﻪدار ﺟﺪاﺷﺪه از ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﭼﺎدرﻳﻨﻪ ﺑﺎ ﺳﻠﻮلﻫﺎي اﺳﺘﺮوﻣﺎل ﻛﺸﺖ ﺷﺪه ﺗﻤﺎﻳﺰ ﻧﻴﺎﻓﺘﻪ ﺟﺪاﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان ﺑﺮ روي رژﻧﺮاﺳﻴﻮن ﻋﺼﺐ ﺳﻴﺎﺗﻴﻚ ﻣﻮرد ﻣﻘﺎﻳﺴﻪ ﻗﺮار ﮔﺮﻓﺖ. ﻃﺮح ﻣﻄﺎﻟﻌﻪ- ﻣﻄﺎﻟﻌﻪ ﺗﺠﺮﺑﻲ درون- زﻳﺴﺘﻲ ﺣﻴﻮاﻧﺎت- 4 ﻗﻄﻌﻪ ﻣﻮش ﺻﺤﺮاﻳﻲ ﻧﮋاد آﻟﺒﻴﻨﻮي ﺳﻔﻴﺪ روش ﻛﺎر- ﻣﻮشﻫﺎي ﺻﺤﺮاﻳﻲ در ﺷﻜﻞ ﺗﺼﺎدﻓﻲ ﺑﻪ ﭼﻬﺎر ﮔﺮوه ﻛﻨﺘﺮل ﻧﺮﻣﺎل، ﮔﺮوه ﮔﺮاﻓﺖ ورﻳﺪ از داﺧﻞ ﺑﺮﮔﺮداﻧﺪه ﺷﺪه، ﮔﺮوه ﺳﻠﻮلﻫﺎي ﻫﺴﺘﻪدار ﺟﺪا ﺷﺪه از ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﭼﺎدرﻳﻨﻪ و ﮔﺮوه ﺳﻠﻮلﻫﺎي اﺳﺘﺮوﻣﺎل ﻛﺸﺖ ﺷﺪه ﺗﻤﺎﻳﺰ ﻧﻴﺎﻓﺘﻪ ﺟﺪاﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان ﺗﻘﺴﻴﻢ ﺷﺪﻧﺪ. ﻣﺤﻞ ﻧﻘﻴﺼﻪ ﻋﺼﺒﻲ ﺑﺎ ﮔﺮاﻓﺖ 10 ﻣﻴﻠﻲﻣﺘﺮي ورﻳﺪ از داﺧﻞ ﺑﺮﮔﺮداﻧﺪه ﺷﺪه ارﺗﺒﺎط داده ﺷﺪ. در ﮔﺮوه ﺳﻠﻮلﻫﺎي ﻫﺴﺘﻪدار ﺟﺪاﺷﺪه از ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﭼﺎدرﻳﻨﻪ و در ﮔﺮوه ﺳﻠﻮلﻫﺎي اﺳﺘﺮوﻣﺎل ﻛﺸﺖ ﺷﺪه ﺗﻤﺎﻳﺰ ﻧﻴﺎﻓﺘﻪ ﺟﺪاﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان ﺑﻪ ﺗﺮﺗﻴﺐ داﺧﻞ ورﻳﺪ ﺑﺎ ﺳﻠﻮلﻫﺎي زﻳﺮﺑﻂ ﭘﺮ ﮔﺮدﻳﺪ. ﻣﻄﺎﻟﻌﺎت ﻋﻤﻠﻜﺮدي ﺿﺮﻳﺐ ﻋﻤﻠﻜﺮد ﺳﻴﺎﺗﻴﻚ، ﻣﻄﺎﻟﻌﺎت اﻟﻜﺘﺮوﻓﻴﺰﻳﻮﻟﻮژﻳﻜﻲ، ارزﻳﺎﺑﻲﻫﺎي رﻳﺨﺖﺷﻨﺎﺳﻲ و اﻳﻤﻮﻧﻮﻫﻴﺴﺘﻮﺷﻴﻤﻲ در ﻫﻔﺘﻪﻫﺎي 16، 20 و 24 ﺑﻌﺪ از ﺟﺮاﺣﻲ ﺑﻪ اﻧﺠﺎم رﺳﻴﺪ. ﻧﺘﺎﻳﺞ- اﺧﺘﻼف ﻣﻌﻨﻲدار(P>0.05) از ﻟﺤﺎظ ﻋﻤﻠﻜﺮد اﻋﺼﺎب رژﻧﺮه، داﻣﻨﻪ و زﻣﺎن ﺗﺎﺧﻴﺮ اﻟﻜﺘﺮوﻣﻴﻮﮔﺮاﻓﻲ در ﻣﻘﺎﻳﺴﻪ دو ﮔﺮوه، ﮔﺮوه ﺳﻠﻮل- ﻫﺎي ﻫﺴﺘﻪدار ﺟﺪاﺷﺪه از ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﭼﺎدرﻳﻨﻪ و ﮔﺮوه ﺳﻠﻮلﻫﺎي اﺳﺘﺮوﻣﺎل ﻛﺸﺖ ﺷﺪه ﺗﻤﺎﻳﺰ ﻧﻴﺎﻓﺘﻪ ﺟﺪاﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان، ﻣﺸﺎﻫﺪه ﻧﺸﺪ. وﻟﻲ در ﻣﻘﺎﻳﺴﻪ ﺑﺎ ﮔﺮوه ﮔﺮاﻓﺖ ورﻳﺪ از داﺧﻞ ﺑﺮﮔﺮداﻧﺪه ﺷﺪه ﻫﺮ دو ﮔﺮوه ﺳﻠﻮلﻫﺎي ﻫﺴﺘﻪدار ﺟﺪاﺷﺪه از ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﭼﺎدرﻳﻨﻪ و ﮔﺮوه ﺳﻠﻮل-ﻫﺎي اﺳﺘﺮوﻣﺎل ﻛﺸﺖ ﺷﺪه ﺗﻤﺎﻳﺰ ﻧﻴﺎﻓﺘﻪ ﺟﺪاﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان اﻓﺰاﻳﺶ رژﻧﺮاﺳﻴﻮن ﻋﺼﺒﻲ ﻳﻜﺴﺎﻧﻲ از ﺧﻮد ﻧﺸﺎن دادﻧﺪ. ﻧﺘﻴﺠﻪﮔﻴﺮي و ﻛﺎرﺑﺮد ﺑﺎﻟﻴﻨﻲ- در ﻣﻄﺎﻟﻌﻪ ﺑﺎﻓﺖ ﺷﻨﺎﺳﻲ روﻧﺪ رژﻧﺮاﺳﻴﻮن ﻋﺼﺐ ﺳﻴﺎﺗﻴﻚ ﻣﻲﺗﻮان ﻧﺘﻴﺠﻪﮔﻴﺮي ﻛﺮد ﻛﻪ ﻧﺘﺎﻳﺞ رﻳﺨﺖﺷﻨﺎﺳﻲ ﻣﻄﺎﻟﻌﻪ ﻃﻮﻻﻧﻲ- ﻣﺪت آن ﺑﻴﺸﺘﺮ ﻗﺎﺑﻞ اﻋﺘﻤﺎد ﺑﺎﺷﺪ.
