Abstract :
Objective- To evaluate growth characteristics and differentiation capacity of equine mesenchymal stem cell (eMSCs) derived from bone marrow (BM).
Study design- In vitro experimental study.
Animals- Four young adult horses (2-5 years old)
Procedure- Cell morphology and growth characteristics of eMSCs harvested from BM were evaluated in standard culture conditions. eMSCs in passage 3 were subjected to osteogenic and adipogenic differentiation induction to investigate their differentiation potential.PCR analysis of differentiated cells was done to determine differentiation.
Results-The cells expanded to sufficient quantity for therapeutic purposes within days and they survived to later passages while sustained their fibroblast-like morphology. Positive osteogenesis was detected via Alizarin Red staining and adipogenesis was confirmed by Oil Red O staining. The cells were more potential to differentiate into osteoblasts rather than adipocytes. PCR analysis approve relative expression of osteogenic and adipogenic genes.
Conclusions and Clinical relevance - The results of this study further support eBM-MSCs as a cell population with the capacity to proliferate and differentiate down the osteogenic and adipogenic lineages. Equine BM is a potentials source of MSCs for cell-based regenerative therapies in horse.

ﻫﺪف- ﺑﺮرﺳﻲ ﺧﺼﻮﺻﻴﺎت رﺷﺪ و ﻗﺎﺑﻠﻴﺖ ﺗﻤﺎﻳﺰ ﺳﻠﻮﻟﻬﺎي ﺑﻨﻴﺎدي اﺳﺘﺨﺮاج ﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان در اﺳﺐ
ﻃﺮح ﻣﻄﺎﻟﻌﻪ- ﻣﻄﺎﻟﻌﻪي ﺗﺠﺮﺑﻲ ﺧﺎرج از ﻣﺤﻴﻂ ﺑﺪن
ﺣﻴﻮاﻧﺎت- ﭼﻬﺎر رأس اﺳﺐ ﺑﺎﻟﻎ ﺟﻮان )ﺳﻦ 2 -5ﺳﺎل(
روشﻛﺎر- ﺳﻠﻮﻟﻬﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﺎل اﺳﺘﺨﺮاج ﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان از ﺟﻬﺖ رﻳﺨﺖ ﺷﻨﺎﺳﻲ و ﺧﺼﻮﺻﻴﺎت رﺷﺪ در ﻣﺤﻴﻂ ﻛﺸﺖ اﺳﺘﺎﻧﺪارد ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﺟﻬﺖ ﺑﺮرﺳﻲ ﻗﺎﺑﻠﻴﺖ ﺗﻤﺎﻳﺰ آدﻳﭙﻮژﻧﻴﻚ و اﺳﺘﺌﻮژﻧﻴﻚ، ﻓﺮاﻳﻨﺪ اﻟﻘﺎء ﺗﻤﺎﻳﺰ در ﻣﻮرد ﺳﻠﻮﻟﻬﺎي ﭘﺎﺳﺎژ ﺳﻮم اﻧﺠﺎم ﺷﺪ. آزﻣﺎﻳﺶ PCRﺳﻠﻮﻟﻬﺎي ﺗﻤﺎﻳﺰ ﻳﺎﻓﺘﻪ ﺑﺮاي ﺗﻌﻴﻴﻦ ﺗﻤﺎﻳﺰ اﻧﺠﺎم ﺷﺪ.
ﻧﺘﺎﻳﺞ- ﻃﻲ ﭼﻨﺪ روز، ﺗﻜﺜﻴﺮ ﺳﻠﻮﻟﻬﺎ ﻣﻨﺠﺮ ﺑﻪ ﺗﻮﻟﻴﺪ ﻣﻘﺎدﻳﺮﻛﺎﻓﻲ ﺳﻠﻮل ﺑﺮاي اﻫﺪاف درﻣﺎﻧﻲ ﺷﺪ. ﺳﻠﻮﻟﻬﺎ ﺣﺘﻲ در ﭘﺎﺳﺎژﻫﺎي ﺑﺎﻻ زﻧﺪه ﻣﺎﻧﺪﻧﺪ و ﺷﻜﻞ ﻓﻴﺒﺮوﺑﻼﺳﺘﻲ ﺧﻮد را ﺣﻔﻆ ﻛﺮدﻧﺪ. ﺗﻤﺎﻳﺰ ﻣﺜﺒﺖ ﺳﻠﻮﻟﻬﺎي ﺑﻨﻴﺎدي ﻣﺮاﻧﺸﻴﻤﻲ ﺑﻪ ﺳﻠﻮﻟﻬﺎي اﺳﺘﺨﻮاﻧﻲ ﺗﻮﺳﻂ رﻧﮓ آﻣﻴﺰي آﻟﻴﺰارﻳﻦ رد و ﺑﺮاي ﺗﺎﻳﻴﺪ ﺗﻤﺎﻳﺰ ﺑﻪ ﺳﻠﻮﻟﻬﺎي ﭼﺮﺑﻲ رﻧﮓ آﻣﻴﺰي اوﻳﻞ رد اﻧﺠﺎم ﺷﺪ. آزﻣﺎﻳﺶ PCR ﻧﻴﺰ ﺑﻴﺎن ﻧﺴﺒﻲ ژنﻫﺎي آدﻳﭙﻮژﻧﻴﻚ و اﺳﺘﺌﻮژﻧﻴﻚ را ﺗﺎﻳﻴﺪ ﻧﻤﻮد. ﻗﺎﺑﻠﻴﺖ ﺳﻠﻮﻟﻬﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﺎل اﺳﺘﺨﺮاج ﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان اﺳﺐ در ﺗﻤﺎﻳﺰ ﺑﻪ ﺑﺎﻓﺖ اﺳﺘﺨﻮاﻧﻲ در ﻣﻘﺎﻳﺴﻪ ﺑﺎ ﺗﻤﺎﻳﺰ ﺑﻪ ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﺑﻴﺸﺘﺮ ﺑﻮد.
ﻧﺘﻴﺠﻪﮔﻴﺮي- ﻧﺘﺎﻳﺞ اﻳﻦ ﻣﻄﺎﻟﻌﻪ ﻧﻴﺰ ﻫﻤﺎﻧﻨﺪ ﻣﻄﺎﻟﻌﺎت ﻗﺒﻠﻲ ﻧﺸﺎن داد ﻛﻪ ﺳﻠﻮﻟﻬﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ اﺳﺘﺨﺮاج ﺷﺪه از ﻣﻐﺰ اﺳﺘﺨﻮان در اﺳﺐ ﺑﻪﻋﻨﻮان ﻳﻚ ﺟﻤﻌﻴﺖ ﺳﻠﻮﻟﻲ ﺑﺎ ﻗﺎﺑﻠﻴﺖ ﺗﻜﺜﻴﺮ و ﺗﻤﺎﻳﺰ ﺑﻪ ردهﻫﺎي اﺳﺘﺌﻮژﻧﻴﻚ و آدﻳﭙﻮژﻧﻴﻚ ﻣﻲﺑﺎﺷﻨﺪ و ﻣﻐﺰ اﺳﺘﺨﻮان اﺳﺐ ﻣﻲ ﺗﻮاﻧﺪ ﺑﻪ ﻋﻨﻮان ﻳﻚ ﻣﻨﺒﻊ ﺑﺎﻟﻘﻮه ﺳﻠﻮﻟﻬﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ ﺑﺮاي درﻣﺎنﻫﺎي ﺑﺎزﺳﺎزي ﻛﻨﻨﺪه ﺑﺎ ﭘﺎﻳﻪي ﺳﻠﻮﻟﻲ در اﺳﺐ ﻣﻮرد اﺳﺘﻔﺎده ﻗﺮار ﮔﻴﺮد.
