Abstract :
Objective- To investigate and compare growth potential as well as aging of mesenchymal stem cells (MSCs) derived from rat bone marrow tissue and adipose tissue (AT) occurred at epicardial and epididymal regions. Design- Experimental study. Animals- 10 Wistar Rats. Procedures- Rat MSCs occurred at bone marrow and epicardial and epididymal AT were isolated and culture expanded through several successive passages. Differentiation potential along bone, cartilage and adipose cell lineages was used to verify MSC identity of the isolated cells, and then the cells were comparatively investigated in terms of their colonogenic ability, population doubling time and growth curve characteristics. Furthermore, the number of senescent cells at different passages was quantified using senescent- associated (SA) ß galactosidase staining method. Results- MSCs from both AT appeared to have more proliferation capacity in culture than those from bone marrow since they exhibited significantly more colony number and shorter PDT value (P<0.05). Epicardial AT-MSCs indicated even more significant proliferation capacity than their epididymal counterparts. With respect to cell aging, marrow-MSCs cultures indicated higher percentages of senescent cells than AT-MSCs (P<0.05). Although the percentages of senescent cells in epididmal AT-MSCs were higher than epicardial AT-MSCs but the difference was not statistically significant. Conclusion and Clinical Relevance- Taken together we concluded that rat epicardial AT-MSCs could be appropriate cells for experimental and preclinical settings since they possess more expansion rate and less percentages of senescent cells in culture.

ﻫﺪف - ﻣﻘﺎﻳﺴﻪ ﭘﺘﺎﻧﺴﻴﻞ ﺗﻜﺜﻴﺮ و ﭘﻴﺮي ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ ﻣﺸﺘﻖ از ﻣﻐﺰ اﺳﺘﺨﻮان و ﺑﺎﻓﺖ ﻫﺎي ﭼﺮﺑﻲ ﻧﺎﺣﻴﻪ اﭘﻴﺪﻳﺪﻳﻢ و اﭘﻲ ﻛﺎردﻳﻮم ﻣﻮش ﺻﺤﺮاﻳﻲ. ﻃﺮح ﻣﻄﺎﻟﻌﻪ - ﻣﻄﺎﻟﻌﻪ ﺗﺠﺮﺑﻲ. ﺣﻴﻮاﻧﺎت - 10 ﻣﻮش ﺻﺤﺮاﻳﻲ ﺑﺎ ﻧﮋاد وﻳﺴﺘﺎر. روش ﻛﺎر - ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎدي از ﺑﺎﻓﺖ ﻣﻔﺰ اﺳﺘﺨﻮان و ﺑﺎﻓﺖ ﭼﺰﺑﻲ واﻗﻊ در ﻧﻮاﺣﻲ اﭘﻴﻜﺎردﻳﻮم و اﭘﻴﺪﻳﺪﻳﻢ ﻣﻮش ﺻﺤﺮاﻳﻲ ﺟﺪا ﺳﺎزي ﺷﺪ و ﺑﺎ اﻧﺠﺎم ﭼﻨﺪ ﭘﺎﺳﺎژ ﺳﻠﻮﻟﻲ ﺗﻜﺜﻴﺮ ﮔﺮدﻳﺪ. ﺑﺮاي ﺗﺎﻳﻴﺪ ﻣﺎﻫﻴﺖ ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ ﺳﻠﻮل ﻫﺎي ﺟﺪا ﺷﺪه، از ﺗﻤﺎﻳﺰ ﺑﻪ ﺳﻪ رده اﺳﺘﺨﻮان، ﻏﻀﺮوف و ﭼﺮﺑﻲ اﺳﺘﻔﺎده ﺷﺪ و ﺳﭙﺲ ﺳﻠﻮل ﻫﺎ از ﻟﺤﺎظ ﻓﻌﺎﻟﻴﺖ ﻛﻠﻮن زاﻳﻲ، ﻣﺪت زﻣﺎن دوﺑﻠﻪ ﺷﺪن ﺟﻤﻌﻴﺖ ﺳﻠﻮﻟﻲ و وﻳﮋﮔﻲ ﻫﺎي ﻣﻨﺤﻨﻲ رﺷﺪ ﻣﻮرد ﻣﻘﺎﻳﺴﻪ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﺑﻪ ﻋﻼوه، ﺗﻌﺪاد ﺳﻠﻮل ﻫﺎي ﭘﻴﺮ ﺑﺎ روش رﻧﮓ آﻣﻴﺰي ﺑﺘﺎ ﮔﺎﻻﻛﺘﻮزﻳﺪاز ﺗﻌﻴﻴﻦ و ﮔﺮوه ﻫﺎ از اﻳﻦ ﻧﻈﺮ ﺑﺎ ﻫﻤﺪﻳﮕﺮ ﻣﻘﺎﻳﺴﻪ ﺷﺪﻧﺪ. ﻧﺘﺎﻳﺞ ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ ﺣﺎﺻﻞ از دو ﺑﺎﻓﺖ ﭼﺮﺑﻲ ﺑﻄﻮر ﻣﻌﻨﻲ داري ﺑﻴﺶ از ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎددي ﻣﻐﺰ اﺳﺘﺨﻮان ﺗﻜﺜﻴﺮ ﺷﺪﻧﺪ ) P<0.05(. ﺳﻠﻮل ﻫﺎي ﺑﺎﻓﺖ ﭼﺮﺑﻲ اﭘﻴﻜﺎردﻳﻮم در ﻣﻘﺎﻳﺴﻪ ﺑﺎ ﺳﻠﻮل ﻫﺎي اﭘﻴﺪﻳﺪﻳﻢ از ﺗﻮان ﺗﻜﺜﻴﺮي ﺑﺎﻻﻳﻲ ﺑﺮﺧﻮردار ﺑﻮدﻧﺪ. در راﺑﻄﻪ ﺑﺎ ﭘﻴﺮ ﺷﺪن ﺳﻠﻮل ﻫﺎ در ﻣﺤﻴﻂ ﻛﺸﺖ، درﺻﺪ ﺳﻠﻮل ﻫﺎي ﭘﻴﺮ در ﻛﺸﺖ ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎدي ﻣﻐﺰ اﺳﺘﺨﻮان ﺑﻴﺶ از دو ﮔﺮوه دﻳﮕﺮ ﺑﻮد) 05.P<0(. اﮔﺮﭼﻪ در ﺻﺪ ﺳﻠﻮل ﻫﺎي ﭘﻴﺮ در ﻛﺸﺖ ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ ﻣﺸﺘﻖ از ﺑﺎﻓﺖ ﭼﺮﺑﻲ اﭘﻴﺪﻳﺪﻳﻢ ﺑﻴﺶ از در ﺻﺪ آﻧﻬﺎ در ﻛﺸﺖ ﺳﻠﻮل ﻫﺎي اﭘﻴﻜﺎردﻳﻮم ﺑﻮد وﻟﻲ ﺗﻔﺎوت از ﻟﺤﺎظ آﻣﺎري ﻣﻌﻨﻲ دار ﻧﺒﻮد. ﻧﺘﻴﺠﻪ ﮔﻴﺮي و ﻛﺎرﺑﺮد ﺑﺎﻟﻴﻨﻲ- روي ﻫﻢ رﻓﺘﻪ ﻣﻲ ﺗﻮان ﻧﺘﻴﺠﻪ ﮔﺮﻓﺖ ﻛﻪ ﺳﻠﻮل ﻫﺎي ﺑﻨﻴﺎدي ﻣﺰاﻧﺸﻴﻤﻲ ﻣﺸﺘﻖ از ﺑﺎﻓﺖ ﭼﺮﺑﻲ اﭘﻴﻜﺎردﻳﻮم ﻣﻮش ﺻﺤﺮاﻳﻲ ﺳﻠﻮل ﻫﺎي ﻣﻨﺎﺳﺒﻲ ﺑﺮاي ﻣﻄﺎﻟﻌﺎت ﺗﺠﺮﺑﻲ و ﭘﻴﺶ ﻛﻠﻴﻨﻴﻜﻲ ﻣﻲ ﺑﺎﺷﻨﺪ زﻳﺮا از ﺗﻮان ﺗﻜﺜﻴﺮ ﺑﺎﻻﻳﻲ ﺑﺮﺧﻮرددار ﻫﺴﺘﻨﺪ و در ﺻﺪ ﺳﻠﻮل ﻫﺎي ﭘﻴﺮ در ﻛﺸﺖ آﻧﻬﺎ ﭘﺎﻳﻴﻦ اﺳﺖ.
