Abstract :
Lentinula edodes and its mycelium are known for their unique properties and have been used in food and medicinal industry for centuries. The effect of different temperatures and medium conditions on yield of dry mycelium and total amino acid content was studied. Submerged culture technique was employed to grow mycelium in PDB (potato dextrose broth) and PDB++ (PDB, yeast extract, malt extract, peptone, KHl04 and MgS0 4.7H20 ) media under different temperatures. The amino acid composition of the mycelium was determined by the High Performance Liquid Chromatography techniques (HPLC). An optimum growth of the mycelium was observed on the PDB++ medium incubated at 25 °C. The mycelium harvested had the highest content of amino acid, of which the major component was glutamic acid.
