Abstract :
An experiment was conducted to steadythe effect of L-Tryptophanand salicylic acid oncallusinducedon leaf explants of Catharanthus roseus. Callus induction was achieved by culturing true leaves of the plant onMurashige and Skoog(MS)mediumsupplemented with 0.5 mg /L 2,4-D and 1mg / L Kin.The best medium to maintain callus was MS medium supplementedwith 0.5 mg /L 2,4-D and 1.5 mg / L Kin. Differentlevels addedto MS medium for each L-Tryptophan0,200,300or400mg /Landsalicylic acid0, 0.5,1or1.5mg /L.The medium supplementedwith 30 g/ L sucrose was used as acontrol. Results showed themedium supplementedwith 200mg L of L-Tryptophan gavethe highest quantity of Vincristinereached48.66μg/100 fresh weight of callus. MSmedium content at the concentration 1mg L of salicylic acidgave the highest level of Vinblastinerecording50.98μg/100 fresh weight ofcallus.While MS medium supplemented with0.5mg /L of salicylic acidgave the highest level of Serpentine24.76μg/100 fresh weight of callus.The concentrations of active compound derived from plantleaves were much less than the concentrations produced by the callus grown in vitro. The concentration of Serpentinewas 0.059whileVinblastinewas0.183andthe concentration of Vincristinewas0.064.
