Abstract :
A sensitive and simple analytical method is described for the electrochemical behaviour and quantitative determination of Josamycin antibiotic drug in its dosage forms and body fluids. The developed AdSV method was based on the adsorptive accumulation of the drug at a hanging mercury drop electrode, followed by the electrochemical reduction process, which yielded a well-defined cathodic peak at - 1.38 V (vs. Ag/AgCl electrode). This electroanalytical response was further optimised with respect to various experimental and operational parameters. A linear calibration graph was obtained in the range 1.5 ×10-7- 3.5 ×10-7moll-1 with a detection limit of 4.3 ×10-8moll-1after 120 s accumulation period at 0.0 V. The reproducibility of the method in terms of RSD% for 10 measurements was 1.8% with a mean recovery of 100.7%. Possible interferences by other pharmaceutical drugs and surface-active materials were also investigated. The proposed procedure was successfully applied for the analysis of the studied drug in commercially available dosage form and in spiked human serum and urine.
