Abstract :
Abstract. The objective of this study was to identify RAPD and STSmarkers linked to stripe rust resistance region Lr46/Yr29 in F3population of wheat by using the bulked segregant analysis method.F3 population has been derived from the cross between the stripe rustresistance parent (monogenic line Lr46/Yr29) and the susceptibleparent (Sids-6) and the evaluation to stripe rust resistance was made atadult stage. Bulk segregant analysis was used in conjunction withRAPD analysis. After Mapmaker linkage analysis on the F3population, a standard maximum likelihood technique was employedto analyze the linkage between Yr29 and the linked marker loci. Thegenetic distance between Lr46/Yr29 gene and RAPD marker (OPB-8280) was 12.8 cM with LOD scores of 6.4. Two STS primers,BE518048 and TC265717 were tested by using different PCRprograms with different annealing temperatures to revealpolymorphism between the monogenic line Lr46/Yr29 and the cultivarSids-6. EST- based STS marker BE518048600 was mapped toLr46/Yr29 gene by using the bulked segregant analysis (BSA)procedure with the same previous population. After Mapmakerlinkage analysis on the F3 population, a standard maximum likelihoodtechnique was employed to analyze the linkage between Yr29 and thelinked marker loci. The genetic distance between Lr46/Yr29 and STSmarker BE518048 600 was 11.3 cM with LOD scores of 7. RAPD andSTS markers results revealed that Sids-6 has not Yr29 which linked to Lr46, when evaluated to stripe rust resistance at adult stage. Theavailability of such RAPD and STS markers should accelerate theintrogression of the resistance gene into current wheat cultivars. It willalso facilitate the accumulation of several resistance genes in a targetcultivar.
