Abstract :
Felty Germander (Teucrium polium L.) is a Jordanian medicinal plant that is widely used in folk medicine. It is an endangered plant due to its increased popularity so it must be highly considered for conservation. Medium term conservation of T. polium was successfully achieved using elevated concentrations of different osmotic agents. Approximately 0.7 cm micro-shoots were aseptically excised from in vitro grown plants and transferred to Hormone Free-MS medium (HF-MS) for 4-5 days. Pre-treated micro-shoots were transferred to MS media plus elevated concentrations of abcisic acid (ABA), mannitol, sorbitol, or sucrose, for 12 weeks. Approximately 33.3% of T. polium micro-shoots were regrown when culturing on MS meidum supplemented with 3.8 ?M ABA. Addition of 0.1 M mannitol to the T. polium conservation medium resulted in 36.3% regrowth, whereas using 0.1 M sorbitol resulted in 57.3% regrowth. Using medium containing 0.2 M sucrose gave 90% regrowth of T. polium micro-shoots.
