Abstract :
Quantitative determination of Fluconazole in IV solutions was carried out by UV spectrophotometric method using the absorbance values at 261.6 nm and the second order derivative absorbance values at 274 nm. Distilled water was used as solvent throughout the study. The concentration range was 4.0 × 10-4-1.5 × 10-3 M for the two spectrophotometric methods. While detection limit was 4.37 µg/mL in direct UV method, it was 2.63 µg/mL in derivative spectrophotometric method. In addition, the slope of the calibration curve in derivative spectrophotometry was higher than the slope obtained by direct UV method. Percent recovery values at 261.6 nm were 97.85% for capsules, 96.95% for IV solutions respectively; but these values were 97.20% for capsules and 104.43% for IV solutions in second order derivative spectrophotometry at 274 nm (n = 5). RSD values at 274 nm in the second order derivative spectrophotometric method were 0.52 for capsules and 0.92 for IV solutions. RSD values showed that both spectrophotometric methods were reproducible. In capsules, the two spectrophotometric methods were performed and found to be comparable to the non-aqueous titrimetric method which was applied by using anhydrous acetic acid as solvent and perchloric acid as titrant. The obtained results were compared to each other.
