Purpoe: To evaluate the effectivene and afety of olopatadine hydrochloride and to determine it optimal concentration and the onet and duration of action for treating allergic conjunctiviti. Olopatadine i a new topical ophthalmic antiallergic agent that demontrate activity a both an antihitamine and a mat cell tabilizer. Two double-maked, randomized, placebo-controlled, contralateral eye comparion tudie were conducted uing the conjunctival allergen challenge model. Method: A total of 169 ubject received 0.05% or 0.1% olopatadine. tudy ubject were healthy adult men and women with a hitory of active allergic conjunctiviti within the previou two eaon but not receiving current treatment. With an allergen doe that produced ign and ymptom of allergic conjunctiviti at viit 1 and 2, the conjunctival allergen challenge wa performed 27 minute after tudy drug adminitration at the third viit (onet-of-action challenge) and at 8 hour after tudy drug adminitration at the fourth viit (duration-of-action challenge). Olopatadine wa adminitered in one eye and placebo in the oppoite eye. Itching and redne were cored for both eye at 3, 10, and 20 minute after the conjunctival allergen challenge. Reult: Both 0.05% and 0.1% concentration of olopatadine were ignificantly (P < .05) more effective than placebo in inhibiting itching and redne at all evaluation when adminitered 27 minute or 8 hour before the conjunctival allergen challenge. There were no eriou or drug-related ocular or nonocular advere event in either tudy. Concluion: Thee finding demontrate the rapid and prolonged (at leat 8 hour) ocular antiallergic action of olopatadine.
