HLA-DRB and HLA-DQB loci in the genetic uceptibility to develop glaucoma in Mexican Original Reearch Article

PURPOE: Glaucoma i a clinically heterogeneou dieae with a pathophyiology that may include genetic uceptibility, poibly aociated with an immunologic diorder. The aim of thi tudy wa to determine whether the DNA polymorphim located in the HLA-DRB1 and HLA-DQB1 gene how a pecific aociation pattern in Mexican metizo patient with primary open-angle glaucoma. METHOD: Thi wa a cro-ectional, cae-control, multicenter tudy. We analyzed the HLA-DRB1 and DQB1 loci of 81 Mexican metizo nonrelated patient with primary open-angle glaucoma and 98 healthy ethnic matched control ubject. Patient were diagnoed clinically and by viual field examination. HLA typing wa performed by PCR-O revere dot blot. REULT: We documented increaed frequencie of HLA-DRB1*0301, DRB1*1101, DRB1*0701, DRB1*1402, DQB1*0302, and DQB1*0301; however, none of them were ignificantly different from normal control ubject. Haplotype analyi howed that the HLA-DRB1*0407-DQB1*0302 haplotype i ignificantly increaed in patient compared with control ubject (P = .0001). CONCLUION: The haplotype HLA-DRB1*0407-DQB1*0302 i common among Mexican metizo (haplotype frequency = 0.102), and it wa increaed in our patient (haplotype frequency = 0.259, P = .0001). Thi may reflect an independent aociation of thi haplotype with the dieae a the reult of linkage diequilibrium or the influence of a neighboring gene. The pathophyiology of thi illne i uncertain, and further tudie are needed regarding the genetic uceptibility to develop primary open-angle glaucoma.
