Abstract
PURPOE: To determine whether equence analyi of 16 riboomal DNA (rDNA) can be ued to detect bacterial pathogen in patient with potoperative endophthalmiti.
METHOD: In 10 eye of 10 patient, vitreou pecimen were collected for culture and rDNA typing. Variable egment of each riboomal DNA pecimen were amplified by polymerae chain reaction (PCR), equenced, and aligned by BLAT, a computer alignment program, againt equence in GenBank at the National Intitute of Health.
REULT: pecimen were available from five eye with bacterial endophthalmiti diagnoed by Gram tain or culture. Amplified 16 rDNA equence from the eye of three patient were identical to microbiologic reult. Polymerae chain reaction reult were negative in two cae in which unuual organim were detected. All five control pecimen from patient with nonbacterial endophthalmiti or uveiti were PCR negative. Approximately 48 to 72 hour are required under ideal condition for final pecie identification with thi riboomal typing technique.
CONCLUION: 16 rDNA typing how potential a a relatively rapid technique for identifying bacteria in vitreou ample.
