Abstract :
PURPOE: To evaluate change in the rod and amacrine cell and horizontal cell in human retina with retiniti pigmentoa.
METHOD: even retina from patient donor with retiniti pigmentoa and 14 age- and potmortem-matched normal human retina were proceed for immunocytochemitry and confocal microcopy. The following cell-pecific antibodie were ued: anti-rhodopin (rod), anti–γ-aminobutyric acid (amacrine cell), anticalbindin (cone and horizontal cell), anti–glial fibrillary acidic protein (atrocyte and reactive Müller cell), and anti-ynaptophyin and anti-V2 (ynaptic veicle).
REULT: In retinal region with ignificant photoreceptor lo, the rod, γ-aminobutyric acid–poitive amacrine cell, and calbindin-poitive horizontal cell had undergone neurite prouting. The rod, amacrine and horizontal cell neurite were aociated with the urface of glial fibrillary acidic protein–immunoreactive Müller cell. Mot rod neurite that projected into the inner retina contacted the omata of γ-aminobutyric acid–poitive amacrine cell.
CONCLUION: Rod, amacrine and horizontal cell undergo neurite prouting in human retina with retiniti pigmentoa. Thee change in the retinal neuron may contribute to the electroretinographic abnormalitie and progreive decline in viion noted by patient with retiniti pigmentoa. Thee alteration may alo complicate trategie for treatment of retiniti pigmentoa.
