Additive efficacy of unoprotone iopropyl 0.12% (recula) to latanoprot 0.005%

PURPOE: To evaluate the afety and efficacy of adding unoprotone iopropyl 0.12% v placebo both given twice daily to latanoprot 0.005% given every evening. METHOD: We treated 41 patient with primary open-angle glaucoma or ocular hypertenion with latanoprot 0.005% for 1 month and then randomized each to either placebo or unoprotone iopropyl 0.12% for 8 week. Diurnal intraocular preure were meaured at 08:00, 10:00, 12:00, 18:00, and 20:00 hour, both at baeline (time of randomization) and after 8 week of treatment. REULT: Twenty patient were treated in the placebo group and 21 in the unoprotone iopropyl group. After 8 week of treatment in the placebo group, the trough intraocular preure at 08:00 and the diurnal preure were 20.4 ± 3.2 and 19.1 ± 2.2 mm Hg, repectively. In the unoprotone iopropyl group the preure were 19.4 ± 3.3 and 18.0 ± 1.7 mm Hg (P = .22 and P = .042), repectively. However, eye with a baeline preure of 22 mm Hg or greater on latanoprot had an average 3.3 mm Hg greater reduction at trough (P < .01) and a 2.1 mm Hg greater decreae in diurnal preure (P = .030) after adding unoprotone iopropyl (n = 14 eye) compared with placebo (n = 16 eye; P < .001). In addition, the range of the preure throughout the diurnal curve wa reduced from 2.7 mm Hg on latanoprot alone to 1.4 mm Hg after adding unoprotone iopropyl. Advere event were imilar between group, and no patient were dicontinued becaue of afety reaon. CONCLUION: Thi tudy ugget that unoprotone iopropyl can afely improve the diurnal curve characteritic in patient who continue to have an elevated preure on latanoprot 0.005% alone.
