Abstract :
PURPOE: To compare the intraocular preure)–lowering effect and ide effect of latanoprot 0.005% once daily with unoprotone 0.12% twice daily. METHOD: ixty patient with primary open-angle glaucoma or ocular hypertenion were randomized to receive either latanoprot once daily in the evening and placebo once daily in the morning, or unoprotone twice daily in the morning and evening. The tudy wa double maked and followed a croover deign with two treatment period of 1 month eparated by a 3-week wahout period. The intraocular preure wa meaured at 9 and 5 on the baeline and day 28 viit, and at 9 on day 2 and day 14 viit of each treatment period. The 9 meaurement wa taken 2 hour and 13 hour after the lat drop of unoprotone and latanoprot, and the 5 meaurement wa at 10 and 21 hour, repectively. The mean of the meaurement wa calculated. afety parameter were alo recorded. REULT: Fifty-ix patient completed both treatment period and had intraocular preure data available for evaluation. After 1 month of treatment, latanoprot ignificantly reduced intraocular preure (mean ± EM) by 6.1 ± 0.5 mm Hg (P < .001) and unoprotone by 4.2 ± 0.4 mm Hg (P < .001) adjuted from an overall baeline of 22.3 ± 0.5 mm Hg and 23.2 ± 0.4 mm Hg, repectively. The difference of 1.9 mm Hg between treatment wa tatitically ignificant in favor of latanoprot [P = .003, analyi of covariance (ANCOVA)]. Unadjuted analyi of reponder uing the percentage decreae in intraocular preure howed that the proportion of reponder in the latanoprot-treated group wa greater than in the unoprotone-treated group. Advere ocular ymptom and finding were mild in both treatment group. Eye redne and ocular irritation were the mot frequently reported event. CONCLUION: Latanoprot once daily wa ignificantly more effective in reducing intraocular preure compared with unoprotone twice daily after 1 month of treatment in patient with primary open-angle glaucoma and ocular hypertenion. Both drug were well tolerated with few ocular advere event.
