Abstract :
PURPOE: To determine the effect of the topical ocular hypotenive drug, iopropyl unoprotone, a docoanoid molecule with very weak protaglandin activity, on herpe keratiti in the rabbit eye.
METHOD: For acute dieae, rabbit cornea inoculated with the corticoteroid-enitive F(MP)E train of herpe implex viru type 1 were treated with variou combination of 0.12% iopropyl unoprotone, latanoprot, trifluridine, benzalkonium chloride 0.02%, dexamethaone odium phophate, ketorolac tromethamine, or aline olution beginning 1 day after infection. everity of keratiti wa evaluated in a maked manner. For recurrent dieae, rabbit cornea infected with McKrae train herpe implex viru type 1 were treated with unoprotone or aline olution on potinfection day 25 to 42, and the preence or abence of leion wa recorded.
REULT: Eye treated with unoprotone howed ignificantly le evere dieae than aline-treated or latanoprot-treated eye during acute infection. Unoprotone-treated and aline-treated eye howed no ignificant difference in the frequency of recurrent leion. Eye treated with latanoprot and/or dexamethaone, eparately or in combination, howed increaed everity of acute herpe implex viru keratiti, wherea benzalkonium chloride 0.02%–treated eye howed no ignificant difference, compared with aline treatment. Trifluridine reulted in rapid healing.
CONCLUION: Unoprotone did not increae the everity or recurrence rate of herpe implex viru keratiti. Unoprotone require twice-a-day adminitration, compared with once-a-day for latanoprot, and unoprotone lower intraocular preure le than latanoprot. Neverthele, unoprotone’ uperior afety profile may make it ue advantageou. Benzalkonium chloride alone did not make the keratiti wore.
