Abstract :
PURPOE: Paraoxonae (E.C.3.1.1.2) i a polymorphic protein hown to prevent low-denity lipoprotein oxidation. Our purpoe i to evaluate the hypothei that paraoxonae gene polymorphim and plama oxidized low-denity lipoprotein level play a role in the occurrence of exudative age-related macular degeneration. METHOD: We analyzed paraoxonae genotype (A/B, Gln-Arg192 and L/M, Leu-Met54) and plama oxidized low-denity lipoprotein level in 72 unrelated Japanee patient with exudative age-related macular degeneration and compared the reult with thoe of 140 age-matched and ex-matched control ubject. REULT: The ditribution of paraoxonae 192 and paraoxonae 54 polymorphim wa ignificantly different between the patient with age-related macular degeneration and control ubject (chi-quare = 6.226, P = .0445, and chi-quare = 6.863, P = .0323, repectively). The high frequency of the BB genotype at poition 192 wa oberved in the exudative age-related macular degeneration group compared with control ubject (52.8% v 35.0%, repectively; P = .0127). The high frequency of the LL genotype at poition 54 wa oberved in the patient more than the control (91.7% v 77.1%, repectively; P = .0090). The mean (± E) oxidized low-denity lipoprotein level in the patient wa ignificantly higher than in the control (19.1 ± 1.0 v 16.2 ± 0.6 U/ml, P < .01). CONCLUION: Thee reult indicate that the paraoxonae gene polymorphim may repreent a poible genetic rik factor for age-related macular degeneration and that increaed plama oxidized low-denity lipoprotein may be involved in the pathogenei of age-related macular degeneration.
