Abstract :
PURPOE: The aim wa to compare topical brinzolamide 1% twice daily with dorzolamide 2% twice daily, each given with timolol 0.5% twice daily, for afety and effect on intraocular preure in patient with primary open-angle glaucoma or ocular hypertenion. METHOD: Thi double-blind, randomized, active controlled, parallel group tudy wa conducted multinationally at 31 ite, in 241 patient a above, with aement at baeline and monthly during 3 month of treatment. The primary end point wa a diurnal reduction of trough/peak intraocular preure from a timolol 0.5% twice daily baeline. REULT: Both treatment regimen reduced intraocular preure ignificantly at all time point (P < .001): brinzolamide plu timolol by −3.6 to −5.3 mm Hg (−14.2 to −21.9%), dorzolamide plu timolol by −3.6 mm Hg to −5.1 mm Hg (−14.1 to −21.2%). Clinically relevant intraocular preure reduction (decreae 5 mm Hg or greater or abolute intraocular preure value 21 mm Hg or le) were manifeted by 50.0% to 89.3% of patient under brinzolamide plu timolol and by 43.9% to 85.4% under dorzolamide plu timolol. The treatment were equivalent in mean intraocular preure-lowering. In general, both regimen were well tolerated. However, more patient (P = .001) experienced at leat one advere event with dorzolamide plu timolol (32.8%) a compared with brinzolamide plu timolol (14.7%); alo, more patient (P = .001) experienced ocular dicomfort (tinging and burning) after dorzolamide plu timolol (13.1%) than after brinzolamide plu timolol (1.7%). CONCLUION: In term of intraocular preure reduction, brinzolamide 1% twice daily wa equivalent to dorzolamide 2% twice daily, each added to timolol 0.5% twice daily, but brinzolamide produced ignificantly le ocular burning and tinging.
