Abstract :
Purpoe To invetigate the preence of the fibronectin ioform containing the extradomain B (B-FN), a marker-protein of angiogenei, in urgically excied human choroidal neovacular membrane (CNVM) to evaluate whether B-FN could be ued a a therapeutic target for pecific antibody-photoenitizer immunoconjugate. Deign Laboratory invetigation. Method The etting wa an intitutional practice. The tudy population conited of 15 eye (15 patient) with CNVM undergoing membrane exciion (four eye with age-related macular degeneration, even with pathologic myopia and four with multifocal choroiditi). The control group conited of eight eye bank eye (four ubject) without choroidal neovacularization. Light microcopic immunohitochemitry on cryotat ection of tiue wa obtained. B-FN wa detected by a human recombinant antibody, CG-1, and compared with immunotaining for endothelial cell with factor VIII–related antigen. The main outcome meaure wa the preence of CG-1 poitively tained cell or area of the extracellular matrix. taining of CG-1 wa cored on a cale from 0 to 3. Reult Fourteen of 15 neovacular membrane tained trongly with CG-1 (core 2 or 3). One membrane from a patient with pathologic myopia wa negatively tained (core 0). CG-1 poitive taining wa detected around endothelial cell and in the extracellular matrix of CNVM. The retina of eye without choroidal neovacularization wa negative with CG-1 in all eight donor eye, while the choroid contained ome weakly CG-1 poitive cell (core 0 and 1, repectively). Concluion The extradomain B i abundantly expreed in CNVM, but it expreion i more retricted in eye harboring no apparent choroidal neovacularization. In the future, B-FN might erve a a target for the delivery of antibody-photoenitizer immunoconjugate to newly developed veel to enhance the electivity of photodynamic therapy.
