Abstract :
Purpoe To examine the expreion of the three interferon-γ-inducible CXCR3-binding chemokine, CXCL10/IP-10 (interferon-γ-inducible protein of 10 KDa), CXCL9/Mig (monokine induced by interferon-γ), and CXCL11/I-TAC (interferon-inducible T-cell α chemoattractant) in the conjunctiva of patient with vernal keratoconjunctiviti (VKC). Thee chemokine exhibit potent T-lymphocyte chemoattractant activity. Deign Immunohitochemical tudy. Method Conjunctival biopy pecimen from 16 patient with active VKC and nine control ubject were tudied by immunohitochemical technique uing monoclonal antibodie directed againt IP-10, Mig, and I-TAC. The phenotype of inflammatory cell expreing chemokine wa examined by double immunohitochemitry. Reult In the normal conjunctiva, very weak Mig immunoreactivity wa oberved on baal epithelial cell and on vacular endothelial cell in the upper ubtantia propria. There wa no immunoreactivity for the other chemokine. In all VKC pecimen, trong immunoreactivity for Mig wa expreed by epithelial cell, vacular endothelial cell, and inflammatory mononuclear cell. Inflammatory mononuclear cell expreing IP-10 and I-TAC were noted in 10 and nine pecimen, repectively. The number of Mig+ inflammatory cell were ignificantly higher than the number of IP-10+ and I-TAC+ inflammatory cell (P < .001). Inflammatory cell expreing Mig were CD4+ T-helper/inducer cell (71.6 ± 3.2%), CD8+ T-cytotoxic/uppreor cell (19.5 ± 1.5%), and CD68+ monocyte/macrophage (5.3 ± 5%). All inflammatory cell expreing IP-10 and I-TAC were CD68+ monocyte/macrophage. Concluion The CXC chemokine Mig i electively and highly expreed in VKC uggeting a pathogenic role of the chemokine receptor CXCR3 and the ligand Mig in the recruitment of activated T lymphocyte.
