Abstract :
Purpoe
Oxidative tre and antioxidant tatu in eye tiue may be aociated with glaucomatou damage. The aim of thi tudy wa to etablih the antioxidant tatu of aqueou humor of patient with primary open-angle glaucoma. For thi purpoe the author meaured the total reactive antioxidant potential (TRAP) and the activitie of the antioxidant enzyme uperoxide dimutae (OD), catalae, and glutathione peroxidae.
Deign
Cae control tudy.
Method
Aqueou humor wa obtained at the time of urgery from 24 patient with glaucoma and 24 cataract patient; TRAP wa meaured by chemiluminecence. Activitie of the antioxidant enzyme were meaured pectrophotometrically. uperoxide dimutae activity wa determined by inhibition of the rate of adrenochrome formation at 480 nm. Catalae activity wa evaluated by decreae of H2O2 aborbance at 240 nm. Glutathione peroxidae (GPx) activity wa determined following nicotinamide adenine dinucleotide phophate oxidation at 340 nm.
Reult
Total reactive antioxidant potential value of the cataract group wa 124 ± 5 μmol/l Trolox. Thi value wa ignificantly decreaed, by 64%, in glaucoma patient. An increae of 57% in OD activity wa oberved in glaucoma patient when compared with cataract patient (41.7 ± 2.7 U OD/ml). Glutathione activity wa threefold higher in glaucoma patient than in the cataract group (6.1 ± 0.6 U/ml). No ignificant change were found in catalae level.
Concluion
Oxidative tre may lead to an induction of antioxidant enzyme and contribute to TRAP decreae. uperoxide dimutae, GPx activitie, and TRAP may be ueful oxidative tre marker in aqueou humor of glaucoma patient.
