Abstract :
Purpoe To examine whether indocyanine green (ICG) dye induce apoptoi in human retinal pigment epithelial (RPE) cell. Deign Laboratory invetigation. Method Pure culture of human RPE cell were iolated. Retinal pigment epithelial cell were incubated with different concentration of ICG dye (1 mg/ml, 5 mg/ml, or 20 mg/ml) for 30 minute. The rate of RPE cell apoptoi wa aeed with Annexin V-FITC taining and propidium iodide (PI) by flow cytometry. Reult Retinal pigment epithelial cell maintained their monolayer morphology after incubation with ICG dye. However, ICG induced a tatitically ignificant amount of apoptoi in RPE cell at all the concentration (1 mg/ml, 5 mg/ml, and 20 mg/ml) after 30 minute of incubation (P< .05). The olvent olution alone (without the ICG dye) did not induce any ignificant apoptoi in RPE cell, when compared with culture medium. Concluion The incubation of RPE cell with ICG dye increaed the number of apoptotic RPE cell in vitro. Our finding indicate that the deciion over the intravitreal application of ICG dye need to be made with caution.
