Trypan blue induce apoptoi in human retinal pigment epithelial cell

Purpoe To examine whether trypan blue dye induce apoptoi in human retinal pigment epithelium cell. Deign Laboratory invetigation. Method Pure culture of human retinal pigment epithelium cell were iolated. The cell were incubated with different concentration of trypan blue (0.5%, 0.10%, and 0.05%) for either 5 or 30 minute. The rate of retinal pigment epithelium cell apoptoi wa aeed with Annexin V-PE taining and flow cytometry. Reult Trypan blue induced a tatitically ignificant amount of apoptoi in retinal pigment epithelium cell at all the concentration (0.5%, 0.10%, and 0.05%) (P < .05). The increae in incubation time (from 5 to 30 minute) led to an increae in the number of apoptotic retinal pigment epithelium cell. Concluion The incubation of retinal pigment epithelium cell with trypan blue increaed the number of apoptotic retinal pigment epithelium cell in vitro. Our reult ugget that deciion regarding the intravitreal application of trypan blue dye need to be made with caution
