Abstract
Purpoe
To tudy the genotype, allelic frequencie, and polymorphim of apolipoprotein E (Apo E) in unrelated Japanee patient with polypoidal choroidal vaculopathy (PCV) or exudative age-related macular degeneration (AMD) and control ubject without macular degeneration.
Deign
Cro-ectional tudy.
Method
Blood ample from 225 ubject older than 50 year were ued. The 225 ubject included 58 patient with PCV, 85 with AMD, and 82 without macular degeneration. Coding exon of the Apo E gene were amplified by polymerae chain reaction, and the DNA equence were determined by direct equencing with an automated equencer.
Reult
Apo E ε3/ε3 wa the mot frequent genotype with a prevalence of 79.3% in PCV patient, 76.5% in AMD patient, and 67.1% in the control ubject. However, the difference in the percentage were not tatitically ignificant among the three group. The mot frequently found allele in the three group wa ε3. Patient with PCV and AMD were le likely to have ε2 and ε4 than the control ubject, but the difference were not tatitically ignificant. Five minor Apo E ingle nucleotide polymorphim, including ε5 and ε7, were found.
Concluion
Japanee patient with PCV and AMD were le likely to have ε2 and ε4 polymorphim, but the difference from the normal were not tatitically ignificant for the Apo E genotype and allelic frequencie.
