Purpoe To report a method for early and correct diagnoi of mycotic keratiti. Deign Clinical laboratory diagnotic tudy. Method Corneal craping of all the four patient were proceed for DNA extraction which were amplified by fungal pecific primer of internal trancribed pacer region I (IT1). Thee product were equenced and analyzed by ingle tranded conformation polymorphim (CP) for pecie identification. Reult The DNA ample from corneal craping of all the four patient were uccefully amplified by the primer pair IT1 and IT2 and imilarity/diimilarity were etablihed by Jaccard’ coefficient. Patient iolate 1 wa identified a Nectria hematococca, iolate 2 a Candida albican, and iolate 3 and 4 were identified a Bipolari papendorfii. Thi led to prompt initiation of antifungal therapy in all the four cae where ueful viion could be retored. Concluion Early and correct diagnoi of mycotic keratiti by polymerae chain reaction could be obtained in all the four cae compared with conventional method, which helped in the prompt initiation of antifungal therapy in patient.
