Abstract :
Purpoe
To determine whether roe bengal and liamine green affect polymerae chain reaction (PCR) detection of herpe implex viru (HV).
Deign
Laboratory invetigation.
Method
Diagnotic corneal craping were evaluated for PCR inhibitory activity. Dacron wab inoculated with roe bengal and liamine green were proceed a clinical ample, inoculated with control HV, varicella zoter (VZV), cytomegaloviru (CMV), and toxoplama DNA and prepared for PCR. The effect of calcium alginate and cotton wab were alo evaluated.
Reult
Roe bengal, liamine green, and calcium alginate not only inhibit PCR detection of HV DNA, but alo detection of VZV, CMV, and toxoplama DNA. Thi inhibition could be overcome by erial dilution and by DNA purification of the ample before PCR.
Concluion
Roe bengal, liamine green, and calcium alginate can inhibit PCR detection of HV DNA. Clinical craping to be ent for PCR diagnotic teting hould be taken before intillation of roe bengal or liamine green.
