Abstract :
Purpoe
To examine change of elect reduction-oxidation (redox) enitive protein from human donor retinal pigment epithelium (RPE) at four tage of age-related macular degeneration (AMD).
Deign
Experimental tudy.
Method
Human donor eye were obtained from the Minneota Lion Eye Bank and graded uing the Minneota Grading ytem (MG) into four tage that correpond to tage defined by the age-related eye dieae tudy (ARED). Protein content in RPE homogenate wa meaured uing Wetern immunoblotting with protein-pecific antibodie.
Reult
The content of everal antioxidant enzyme and pecific protein that facilitate refolding or degradation of oxidatively damaged protein increaed ignificantly in MG tage 3. Thee protein are involved in the primary (copper-zinc uperoxide dimutae [CuZnOD], manganee uperoxide dimutae [MnOD], and catalae) and econdary (heat hock protein [HP] 27, HP 90, and proteaome) defene againt oxidative damage. Additionally, the inulin pro-urvival receptor exhibited dieae-related upregulation.
Concluion
The pattern of protein change identified in human donor tiue graded uing the MG upport the role of oxidative mechanim in the pathogenei and progreion of AMD. The MG ue nearly identical clinical definition and grading criteria of AMD that are ued in the ARED, o our reult apply to clinical and epidemiologic tudie uing imilar definition. Reult from our protein analyi of human donor tiue help to explain altered oxidative tre regulation and cell-urvival pathway that occur in progreive tage of AMD.
