Abstract :
Purpoe
To examine the ultratructural effect of intravitreal bevacizumab on primate eye with particular focu et on the choriocapillari and to examine the influence of vacular endothelial growth factor (VEGF) inhibition on endothelial cell fenetration.
Deign
Animal tudy.
Method
Four Cynomolgu monkey received an intravitreal injection of 1.25 mg bevacizumab. The eye were enucleated and prepared for light and electron microcopy on day one, four, even, and 14. Control eye remained untreated. Choriocapillari endothelial cell fenetration were quantified.
Reult
Choriocapillari endothelial cell fenetration were ignificantly reduced after intravitreal injection of bevacizumab. Fenetration wa lowet on day four (15.9 ± 6.7 per 25 μm) and increaed again from day even to 14, but wa till ignificantly lower than in the control (66.2 ± 9.5 per 25 μm). Denely packed thrombocyte and leukocyte regionally occluded the choriocapillari lumen of treated eye. On day one an increaed number of leukocyte filled in the choriocapillari lumen. Photoreceptor were damaged in two of 40 light microcopic ection. On day one to even, choroidal melanocyte contained giant melanoome. None of thee decribed feature wa found in control.
Concluion
Intravitreal bevacizumab caue ultratructural change in the choriocapillari of primate eye. A ignificant reduction of choriocapillari endothelial cell fenetration i een a early a 24 hour after injection and their number increae again after two week. Thee finding may play a role in the early clinical effect of intravitreal bevacizumab for macular edema. Becaue an increaed rik of circulation diturbance in the choriocapillari cannot be excluded, patient hould be carefully monitored.
