Abstract :
Purpoe To invetigate CC chemokine receptor 4 (CCR4) and CC chemokine receptor 5 (CCR5) expreion, known to be related to the Th2 and Th1 inflammatory pathway, repectively, and human leukocyte antigen-D related (HLA-DR) antigen a hallmark for ocular urface inflammation in patient with uveiti uing conjunctival impreion cytologic pecimen. Deign Cae-controlled tudy. Method Conjunctival impreion cytologic pecimen were obtained from patient with anterior uveiti (n = 26), and their inflammatory profile wa compared with thoe of patient with vernal keratoconjunctiviti (VKC; n = 24), keratoconjunctiviti icca (KC; n = 17), and normal ubject (n = 17). Expreion of CCR4, CCR5, and HLA-DR were analyzed uing flow cytometry and were expreed by determining the percentage of cell expreing the marker in the conjunctival epithelium. Reult CCR4 wa overexpreed in the uveiti group (mean, 19.8% ± 19.7% of poitive cell) and in the VKC group (24.7% ± 20.1%). CCR5 wa expreed only weakly in uveiti patient (6.4% ± 13.1%) and in the normal ubject (2.4% ± 2.4%). HLA-DR expreion by conjunctival cell wa increaed in the uveiti patient (57.4% ± 21.1%) and in the KC group (52.4% ± 12.1%) compared with the VKC group (23.9% ± 26.8%; P < .001) and normal ubject (22.1% ± 19.1%; P < .001). Concluion CCR4, claically related to the Th2 ytem, and HLA-DR both were overexpreed by the conjunctival epithelium in uveiti patient, wherea CCR5, related to the Th1 ytem, wa expreed weakly in uveiti patient. Thee preliminary reult eem to ugget an involvement of the Th2 ytem on the ocular urface in uveiti. Exploration of the ocular urface in uveiti may repreent a new way to undertand better the immune pathway involved in thi complex dieae.
