Abstract :
ABSTRACT: Fusarium ear rot (scab) is one of the most important disease in wheat and other small grain crops, which influenced strongly by weather conditions. The purpose of this study was investigation of changes in oxalate oxidase (OXO) and B-(1, 3)-glucanase transcripts to determine the initiation time point of wheat defense response to Fusarium graminearum. For this purpose, two wheat cultivars including Falat and Sumi3 as susceptible and tolerant cultivar respectively were planted in field at Gorgan agricultural research station. Total RNA was extracted from infected spikes sampled at 1, 3 and 7 days after inoculation. First standard cDNA was synthesized by using total RNA and used as template in quantitative real-time PCR (qRT-PCR). The result showed that the expression of OXO gene in susceptible variety (Falat) was decreased at all time points after inoculation. But the expression of this gene in tolerant variety increased in 7 days after inoculation however it was decreased at 1, 3 days after inoculation. The expression of B -1, 3 glucanase gene in tolerant variety increased in 7 days after inoculation but in susceptible variety, the expression of this gene was much lower than tolerant one at this ) time.
