Abstract :
The medium-term oral carcinogenesis assay is a useful model for studying oral squamous cell carcinomas phase by phase. Based on the multi-step process of carcinogenesis characterized by initiation, promotion, and tumor progression, chronic administration of 4NQO in drinking water simulates rat tongue carcinogenesis similar to its human counterpart. The aim of this article is to show data generated from 4NQO induced-rat tongue carcinogenesis by our research group. In particular, we reviewed studies demonstrating the role of DNA damage, mutagenesis, proliferation status, apoptosis dysregulation, tumor supressor genes expression, xenobiotics metabolizing enzymes and Wnt/beta catenin signalling pathway, and additionally recent evidences on the influence of angiogenesis and metastasis following experimental oral carcinogenesis. Taken together, these data have revealed as relevant biomarkers for understading oral cancer pathogenesis.
