Abstract :
ABSTRACT: To determine contamination of certified barley loads 50 seed samples from Semnan,Yazd,Zanjan,Gom,Ardabil,Khuzestan,Isfahan,Hamedan,golestan and Mazandaran provinces were taken based on ISTA rules in 2010-2011. 400 seeds of each sample were planted on water- soaked blotters ( filter paper) and incubated 24 hours in freezer and for 7-10 days at 20 ?C under 12 hour alternating cycles of NUV light and darkness and then fungi developed on each seed were examined under microscope and identified (Mathur & Kongsdales , 2003). Fungi in the embryos, endosperms and seed coats of the kernels (healthy and black pointed) separately, were determined by plating them on agar medium .The isolated fungi and infection rate indicated that Alternaria alternata, Bipolaris sorokiniana and Fusarium spp by 47.35, 34.68 and 5.63 % were the predominant fungi respectively, followed by Cladosporium Rhizopus, Curvularia, Aspergillus, Stemphylium Penicillium and Chaetomium were of minor importance. The location of fungi mentioned on various parts of seed such as embryos, endosperm and seed coat was determined. Alternaria alternata was isolated from the endosperm and seed coat especially, Fusarium spp from endosperm and others from seed coat. Such attempt on seed mycoflour is necessary for national seed health standards.
