Abstract :
The peresent study describes the procedure for direct shoot regeneration of Capsicum annuum L. using hypocotyls, cotyledon and shoot tip of pepper from in vitro germinated plants on MS medium supplemented with different concentration and combination of auxins and cytokinins. All the experiments were repeated three times. Data on adventitious structures and plant regeneration were statistically analyzed using a completely randomized design. The best shoot regeneration was observed on MS medium cantaining 5 mg/L BAP along with 0.5 mg/L IAA in three types explants. However, when bud were transferred onto elongation medium (MS+BAP+IAA; MS+BAP+IAA+GA3 or MS+BAP+IAA+PAA), these buds frequently were developed into leaf-like feature rather than into normal shoots. Among the tested hormonal regims, combination of GA3 along with IAA and combination of PAA along with IAA were resulted in best response in hypocotyl and shoot tip explants, respectively. Interestingly, this response was observed when the shoot buds were cultured on medium along with activated charcoal. Regeneration shoots rooted in same elongation medium. Then, the complete plantlets with well-developed root and shoot systems were transferred to field condition where they grew to maturity, flowered and fruited normally.
