  • Abstract
Postnatal stem cells free from ethical issues have been proved to be a rich source of pluripotent stem cells. Among the transcription factors that are expressed at high levels in embryonic cells, Sox-2 (sex determining region Y- box-2) have found to maintained pluripotency of the inner cell mass. We studied the expression of Sox-2 transcription factor in Umbilical Cord Blood (UCB) cells of buffalo. Cells were isolated from buffalo umbilical cord and total RNA was isolated. RT-PCR amplicon was sequenced and compared with known sequences in other organisms and revealed high expression of sox-2 in buffalo UCB cells, which suggests UCB cells have properties of primitive pluripotent stem cells. This is the first report on expression profiling of pluripotency determining stem cell markers in umbilical cord blood of buffalo which can be highly valuable in the wound healing and rapid multiplication of elite high productive varieties.
