Management of genetic diversity in organisms requires evaluation of the genetic structure and segregation of resources of the specified species. The present study was conducted to evaluate the genetic diversity and kinship relationship of rainbow trout pre-breeders (Oncorhynchus mykiss) production center Specific Pathogen Free (SPF) rainbow trout in the country using molecular methods. For this purpose, 7 genetic groups of rainbow trout from selected farms and center of the Veterinary Organization and the Iran Fisheries Organization in the provinces of Mazandaran, West Azerbaijan, Kohgiluyeh and Boyer-Ahmad were transferred to the pre- quarantine hall at the Cold-water Fishes Research Center in Tonekabon. After the adaptation period and tagging, the weight and length of the fish were measured in summer 2017 and winter 2018. Then about 3-5 g of the soft and fresh tissue of the end of the male and female breeders' fins of each farm was fixed in 96° ethanol alcohol and transferred to the Genetics Laboratory of the Cold-water Fishes Research Center in Tonekabon. Genomic DNA extraction was performed using ammonium acetate method and polymerase chain reaction was performed using 7 pairs of microsatellite primers.Vertical electrophoresis of 6% polyacrylamide gel and silver nitrate staining were used to observe the alleles and determine their size. The gel images were recorded by a gel documenting device and the bands were scored using INTAS GEL DOC Ver. 0.2.14.0 software. Based on the results, the highest number of real alleles (10.71) and the amount of observed heterozygocytosis (0.49±0.11) was observed in the Fakhari farm fish and the lowest number of real alleles (5.29±0.01) and the amount of observed heterozygocytosis (0.34±0.08) was observed in the Maroufi farm fish. The highest genetic distance (0.09) and the lowest genetic similarity (0.91) were calculated between the (Fakhari and Ghorbani) Farms and the lowest genetic distance (0.02) and the highest genetic similarity (0.98) were calculated between the (Malekitabar and Sarshar) and the (Maroufi and Hadidi) farms. The highest rate of genetic differentiation (0.084) was calculated between the (Ghorbani and Fakhari) Farms and the lowest rate of genetic differentiation (0.014) was calculated between the (Maroufi and Sarshar) Farms. Also the results of analysis of molecular variance showed that the difference in genetic diversity between populations was significant (P<0.001). The results of this study can be used in the management of breeding reserves and programs for selection and breeding of rainbow trout in Iran.
